Yes, on one of my typical machines I install nvidia-glx (usually along with nvidia-kernel-...-amd64 rather than nvidia-kernel-dkms, but that shouldn't matter) and it works fine on reboot. (I too run startx from one of the VTs, except it's usually awesome instead.)
An important thing to look out for is that the nvidia kernel module is loaded on boot. If you're usually used to a high-resolution framebuffer VT when you boot, you'll notice the difference when the nvidia module is loaded because you'll be stuck at a lower resolution.
