I have no experience using Knoppix from a USB stick, but have you
tried using the sudo command with your editor of choice to edit the
file in question?
e.g.,
sudo nano /path/to/file
If the file is still read-only, you might need to do a micro remaster
like I have to to make DVDs that boot in Adriane mode. Sadly, I know
of no simpler method to do this than mount the iso, copy its contents
to a directory on the hard drive, edit files as needed, and then build
a new bootable iso, the first step of which I should probably write a
script for since I mount isos so infrequently I forget the syntax to
do so, and I've already created a script to rebuild the iso since I do
it so infrequently and genisoimage has such complicated syntax.
