If you look at the code in question (you can grab it with apt-get source
user-setup) it's code in debian-installer to setup an install without a
root user.  This line is only added when this is the case.

Yup, that's what the README in xfce4-session recommends but given that
the above is added by user-setup during an install we don't need
anything else in sudoers.

It's a variable in that script.  It's pointing to the target install
(normally /target I think on d-i).
