I did not succeed in finding out the exact syntax for this file.  
Apparently, the file is the result of a concatenation of two files with
different formats.

The header states that lines should be like

alias-name	target-name

but it also contains lines of the form 

alias-name:	target-name

According to my experiments, the first form is ignored, while the
second works smoothly: adding

en_US.ISO-8859-15: en_US.ISO8859-15

allows me to set LANG=en_US.ISO-8859-15 and launch xterm with Xlib
honouring my locale. (see #251000, and possibly merge)

This is deliberate.  I can't find the reference right now, but I think
this is provided for the sake of old X library implementations that did
not use a colon as a field delimiter.

That is in concordance with my understanding the syntax used by modern
Xlib implementations.

Okay.

There is no syntax error, so I am resolving this report.

I will look into adding a shell script to the xfree86 source package
that can be used to update the diff of the locale aliases file for
parity with glibc.
