Edit (as root) the sources.list

# nano /etc/apt/sources.list

Comment out the cdrom entry[*1]
Then update the database:-

# apt-get update

[*1] so it looks like this (example):-
# deb cdrom:[Debian GNU/Linux 6.0.0 _Squeeze_ blah]/ squeeze main
deb http://where_ever/debian/ squeeze main
