You can use lsof to see what processes are using the device, e.g.
antgel $ sudo lsof /dev/dsp
COMMAND PID USER FD TYPE DEVICE SIZE NODE NAME
esd 1026 antgel 5w CHR 14,3 2343954 /dev/dsp0
antgel $
