I tried to start tlmgr in four different ways.
 - as user
   1. update --all
   2. --gui
 - as root
   3. update --all
   4. --gui

It doesn because of "cannot setup TLPDB".

Don't forget to also include minimal examples of other files that are 
needed, e.g. bibtex databases. Often it also helps
to include the logfile. Please, never send included pictures!

If your example file isn't short or produces more than one page of
output (except when multiple pages are needed to show the problem),

Please read the start of the tlmgr call

(running on Debian, switching to user mode!)
...

And then read the section
	USER MODE
in the manual of tlmgr (tlmgr --help)

Basically, usermode works on a private dir (defualt TEXMFHOME) where
you can installl single packages.

The whole of the TL/Debian installation *cannot* be managed with
tlmgr, thus update --all etc is not available unless you 
set up user mode first.
