by default yes, but you can use other types of filesystems, see man

if you build lenny images with the lenny live-helper version on a
squeeze host system, it will just work.
if you want to build squeeze images with lennys version of live-helper,
it will probably not work and you'll suffer from many bugs we fixed since.

since nobodys doing that, probably nobody really knows. if you insist on
knowing it, try it yourself, but don't expect any help with it.
live-helper is dead.
