It should be in /etc/profile or ~/.profile (just for the current user).

I have in /etc/profile

export CLASSPATH=/usr/java/jdk1.5.0_06/lib/tools.jar:/usr/java/jdk1.5.0_06/jre/lib/rt.jar:./
