First question is why? What do you want from a new distro and why can't
Debian Squeeze do it?

Note: you should also look at using a later release as squeeze is now
oldstable and will be dropped from the archives soon once we start the
process for Debian Jessie. Debian Wheezy 7.5 is the current stable.

Generally, you just tweak the existing distro by changing some defaults
or changing the package selection, adding some metapackages to change
what gets installed by default.

Most of the time, you don't actually need to change much.
