No, in my testing it takes 0.2sec. So not really "long time", but
too long for systemd to notice the pidfile at the first try.

That's debian/patches/pidfile. I've tried reverting that patch, and
then systemd finds the pidfile.

Here's some timing info and the exact error message:

Jul 03 21:39:57.509264 sxl systemd[1]: Starting Software speech output for Speakup...
Jul 03 21:39:57.523409 sxl systemd[1]: espeakup.service: PID file /run/espeakup.pid not readable (yet?) after start: No such file or directory
Jul 03 21:39:57.773883 sxl systemd[1]: Started Software speech output for Speakup.

In the changelog, it's this entry:

espeakup (1:0.71-3) unstable; urgency=low
  [...]
  * espeakup.c: create pid file after espeakup is ready only.
