Yes.

The reconfigure call will update the /etc/locale.gen file to uncomment
the wanted locales and run the 'locale-gen' program, which will generate
the needed locale files, while editing /etc/default/locale will change
the default set in the environment.

You can use the 'locale' program to see what locales are active, and
'locale charmap' to check the charset in effect.
